Real-world clinical multi-omics analyses reveal evolutionary trajectories for drug resistance in non-small cell lung cancer
Up to 85% of lung cancer patients have non-small cell lung cancer (NSCLC). Standard of care for NSCLC includes immune checkpoint inhibitors (ICI) and/or platinum-based chemotherapy (CT). However, therapy success is limited and up to 70% of patients are not responding to ICI. Using Tempus Labs' de-identified clinical molecular and response data, we aimed to identify molecular features associated with drug resistance in advanced NSCLC treated with ICI, ICI+CT, or CT, analyzing multi-omics profiling data from pre- and on/post-treatment tumor biopsies from 1,135 patients. Our study reveals distinct molecular characteristics including tumor immune components underlying evolutionary trajectories for drug resistance and provided a list of biomarkers of therapy response.
